Mineralocorticoid Receptor Mediates Cortisol Regulation of Ionocyte Development in Tilapia (Oreochromis mossambicus)

چکیده

Cortisol is the predominant corticosteroid in ray-finned fish since it does not possess aldosterone synthase necessary to produce specific mineralocorticoids. traditionally believed function as a mineralocorticoid. However, effects of cortisol are mediated through receptors other vertebrates, and there an ongoing debate about whether acts glucocorticoid receptor (GR) or mineralocorticoid (MR) teleosts. To investigate this issue, we conducted study using euryhaline Mozambique tilapia (Oreochromis mossambicus) experimental species. The experiment was designed effect on ionocyte development at both cellular gene expression levels tilapia. We administered exogenous antagonists, used immunohistochemistry quantify numbers, performed real-time PCR assess differentiation factor tumor protein 63 (P63) mRNA, epidermal stem cell marker. observed that increased number Na+-K+-ATPase (NKA)-immunoactive ionocytes (increased by 1.6-fold) promoted P63 mRNA 1.4-fold). Furthermore, found addition antagonist Spironolactone inhibited increase (decreased level control group) suppressed (similarly decreased group). also provided evidence for gr, mr, p63 localization cells. At transcript level, mr ubiquitously expressed gill sections present cells (cells labeled with p63), supporting antagonism functional assay results larvae. Our confirmed stimulates MR, rather than GR. Therefore, provide new direction investigating dual action osmotic regulation skin/gill epithelial tilapia, which could help resolve previously inconsistent conflicting findings.

Glucocorticoid Receptor, but Not Mineralocorticoid Receptor, Mediates Cortisol Regulation of Epidermal Ionocyte Development and Ion Transport in Zebrafish (Danio Rerio)

Cortisol is the major endogenous glucocorticoid (GC) both in human and fish, mediated by corticosteroid receptors. Due to the absence of aldosterone production in teleost fish, cortisol is also traditionally accepted to function as mineralocorticoid (MC); but whether it acts through the glucocorticoid receptor (GR) or the mineralocorticoid receptor (MR) remains a subject of debate. Here, we use...

regulated by, and suited to, the environment in which they reproduce. In many teleosts, which spawn in either freshwater or seawater, sperm motility is initiated by osmotic shock when sperm are ejaculated. These osmolality dependent regulatory systems are quite different in freshwater cyprinid teleosts, such as carp, gold fish and zebrafish, and marine teleosts, such as the pufferfish and floun...

Effects of cortisol on ion regulation in developing tilapia (Oreochromis mossambicus) larvae on seawater adaptation.

The yolk diameter of cortisol-treated tilapia (Oreochromis mossambicus) larvae, immersed in freshwater (FW) containing 10 mg L-1 cortisol from 48 h postfertilization to 12 d posthatching, was significantly larger than that of control larvae after 8 d of treatment, suggesting that inhibition on larval growth occurred only after a long-term treatment with cortisol. Tilapia embryos or larvae treat...

Tgm1-like transglutaminases in tilapia (Oreochromis mossambicus)

Among the adaptations of aquatic species during evolution of terrestrial tetrapods was the development of an epidermis preventing desiccation. In present day mammals, keratinocytes of the epidermis, using a membrane-bound transglutaminase (Tgm1), accomplish this function by synthesizing a scaffold of cross-linked protein to which a lipid envelope is attached. This study characterizes the abilit...

Estradiol impairs hyposmoregulatory capacity in the euryhaline tilapia, Oreochromis mossambicus.

Freshwater (FW)-adapted tilapia (Oreochromis mossambicus) were treated with estradiol (E(2)) for 4 days to stimulate protein synthesis and sampled at 0, 4, and 24 h after exposure to 50% seawater (SW). E(2) increased circulating vitellogenin (VTG) levels in large amounts, indicative of unusually high rates of hepatic protein synthesis. E(2) treatment prevented the recovery of plasma osmolality ...
